Tämä TRIM31 kuuluu alaryhmään C V ja siinä näyttää olevan konservatiivisistä domeeneista RING ja B-box2 sekä Coiled. Coil alue (C-C). (Samanlaista CV-ryhmän domaanirakennetta on myös TRIM 40, 56,73 ja 74).Tälla geenillä on havaittu noussutta ilmenemää eräissä tuumoreissa ja se saattaa olla solukasvun negatiivinen säätelijä. Alternatiivisella pleissauksella tulee useita transkriptivariantteja. Geeniä esiintyy paksusuolessa, virtsarakossa ja hieman myös 5 muussa kudoksessa ( nämä ovat pohjukaissuoli, ohutsuoli, mahalaukku, umpisuoli ja testis). Geenin alternatiivi nimi HCG1 tulee sanoista Hemochromatosis Candidate Gene I. TRIM31 sijoittuu sytoplasmisesti ja joltain fraktiolta mahdollisesti assosioituu mitokondriaan. TRIM31 pitoisuuden säätyminen on monimutkainen. Sitä on havaittu kohonneena kroonisessa gastriitissa ja mahasyövässä. Se voinee olla gastrisen karsinogeneesin alkuvaiheen merkitsijä. Keuhkosyöpäsolututkimuksissa TRIM31 plasmiditransfektio oli tuumorin kasvua vaimentava , solusyklin säätelijäsykliini D1 ja sykliiniE vähenivät. Pääteltiin, että TRIM31 voi olla tuumorisuppressiivinen NSCLC-keuhkosyövässä. Tämä TRIM on antiretroviraali. Rakenteessa on 425 aminohappoa. Rakenteesta on tarkka selvitys alla. TRIM31 is downregulated in non-small cell lung cancer and serves as a potential tumor suppressor. Li H, et al. Tumour Biol, 2014 Jun. PMID 24566900 TRIM31 expression was lower in lung cancer cell lines than normal bronchial cell line HBE. Transfection of TRIM31 plasmid was performed in H157 and H1299 cells. TRIM31 overexpression inhibited cell growth rate and colony formation ability in both cell lines. In addition, expression of cell cycle regulator cyclin D1 and cyclin E were decreased after TRIM31 transfection. In conclusion, TRIM31 might serve as a tumor suppressor in non-small cell lung cancer.

Characterization of TRIM31, upregulated in gastric adenocarcinoma, as a novel RBCC protein. Sugiura T, et al. J Cell Biochem, 2008 Nov 1. PMID 18773414 To explore the molecules associated with gastric adenocarcinoma, we used the gene expression profile database of various human tissues and identified TRIM31 upregulated in both patients with chronic gastritis and stomach cancer. TRIM31 is a new member of RBCC proteins composed of RING finger, B-box and coiled-coil domains. We characterized TRIM31 biochemically and found it possess properties in common with other RBCC proteins, such as occurrence of alternative splicing transcripts, in vitro autoubiquitylating activity and a tendency to homo-oligomerize. The primary localization site of TRIM31 is the cytoplasm but some fraction is potentially associated with the mitochondria. TRIM31 overexpression suppresses colony formation of HCT116 cells while knockdown of its expression with short interfering RNAs (siRNAs) consistently tends to enhance growth of AsPC-1 cells slightly. Thus, TRIM31 is a characteristic RBCC protein with the ability to regulate cell proliferation negatively and may be a potential biomarker of gastric cancer as it is overexpressed from the early stage of gastric carcinogenesis.

(Hemochromatosis Candidate Gene).
