MUC2 11p15.5. Suoliston ja keuhkoputken musiini-2 ( Crohnin tauti, UC ja IBD)

(Finnish)  Yhteenvetoa tästä geenistä MUC2 , joka koodautuu kromosomikohdasta 11p15.5.
Geenillä on vaihtoehtoisia nimiä MLP, SMUC, MUC-2 . MLP viittaa sanoihin   "human intestinal  mucin-like protein (1992)",  suoliston musiininkaltainen proteiini.
  Musiinit ovat  suurimolekyylipainoisia glykoproteiineja, joita monet epiteelikudokset tuottavat, kuten allaolevasta MUC-2- prekursorista näkyy:  sillä on  prekursoripeptidissään  5239  aminohapon sekvenssi. Tämä proteiini  muodostaa normaalisti  läpäisemättömän mukoosiesteen, joka suojaa terveen suoliontelon sisäpintoja.  Proteiini pystyy polymerisoitumaan geelimuotoon, joka koostuu 80%:sti  oligosakkaridipitoisista sivuketjuista,  joista muodostuu  proteiinin suuri molekyylipaino. Peptidi sekvenssistä huomaa, että siinä on toistojaksoja (TR, tandem repeat), joita treoniinit ja proliinit muodostavat (T ja P).  Ne  ovat vaihtelevia  ja niitä on 50-110  kopiota  eri yksilöillä. ( Alla olen  merkannut selvensseissä    mustalla vain  5 peräkkäistä  treoniinia (TTTTT) , mikä helpottaa havaitsemaan erilaisten toistojaksojen esiintymisiä  "peptidikankaankudonnassa") . Tämä  MUC2-geeni säätyy alas  Crohnin tautia ja ulseratiivisa koliittia (UC) potevilla henkilöillä.

  • Summary: This gene encodes a member of the mucin protein family. Mucins are high molecular weight glycoproteins produced by many epithelial tissues. The protein encoded by this gene is secreted and forms an insoluble mucous barrier that protects the gut lumen. The protein polymerizes into a gel of which 80% is composed of oligosaccharide side chains by weight. The protein features a central domain containing tandem repeats (TR) rich in threonine (T) and proline (P) that varies between 50 and 115 copies in different individuals. Downregulation of this gene has been observed in patients with Crohn disease and ulcerative colitis (UC). [provided by RefSeq, Oct 2016]

    Referenssi 1(2018) . Helicobakteerin virulenssitekijän CagA ja ammonium jonin vaikutukset musiineihin AGS- soluissa. H. pylori saattaa vaikuttaa MUC5AC, MUC2, MUC5B ja MUC6 - musiinien ilmeentymiseen AGS- soluissa  virulenssitekijän CagA ja /tai NH:n kautta, mutta ei E-cadheriiniväliteisesti.

    Referenssi 2 (2018): Hirschsprungin  taudissa (HSCR) on goblet-solujen funktio muuntunut. Tämä  on ensimmäinen raportti  TFF-, SPDEF-, KLF4-  ja goblet-soupopulaation ilmenemän vähentymisestä  HSCR- tautia potevilla.  muuntunut goblet-solufunktio voi johtaa suoliston mukosaesteen  dysfunktioon, mikä antaa osaltaan vaikutuksensa HAEC:n kehittymiseen.

MUC2 ilmentymän suolistospesififisyys tunnistettiin hiirestä, ihmisestä ja eedelleen varmistettiin  kananpojjilla.
